چکیده

Epithelial cadherin (E-cadherin) is a transmembrane receptor that plays a vital role in Ca2+-dependent homotypic cell–cell adhesion processes (Takeichi, 1991). The extracellular domain of E-cadherin contains five tandem repeats of about 100 amino acid residues, and is responsible for recognizing cadherin molecules of the same type in a calcium-dependent manner (for a review, see Alattia et al., 1999). The determination of the three-dimensional structure and dynamics of the cadherin extracellular domain in the presence and in the absence of Ca2+ should help us understand how cadherin interacts with Ca2+ ions and mediates cell adhesion functions.

Forced unfolding modulated by disulfide bonds in the Ig domains of a cell adhesion molecule.

Cell adhesion molecules (CAMs) mediate cell attachment and stress transfer through extracellular domains. Here we forcibly unfold the Ig domains of a prototypical Ig superfamily CAM that contains intradomain disulfide bonds. The Ig domains of all such CAMs have conformations homologous to cadherin extracellular domains, titin Ig-type domains, and fibronectin type-III (FNIII) domains. Atomic for...

E-cadherin Promoter Methylation Comparison and Correlation with the Pathological Features of the Squamous Cell Carcinoma of Esophagus in the High Risk Region

E-cadherin is among tumor suppressor genes which mostly subjects to the down-regulation in squamous cell carcinoma of esophagus (SCCE). The gene is tightly associated with the tumor invasion and metastasis in multiple human cancers, especially SCCE. CpG islands’ methylation in the promoter region of E-cadherin is among the mechanisms that have been suggested for the E-cadherin silencing, howeve...

Motifs of cadherin- and fibronectin type III-related sequences and evolution of the receptor-type-protein tyrosine kinases: sequence similarity between proto-oncogene ret and cadherin family.

Immunoglobulin (Ig)-, fibronectin type III (FN-III)-, and cadherin-related sequences are often found in multiple repeats in the extracellular regions of various cell adhesion molecules. The amino acid sequences of 82 different cadherin-like repeats from 13 known members of the cadherin superfamily were compared for six highly conserved regions, and a frequency matrix represented as amino acids ...

P-37: The Effect of Extracellular Calcium Concentration on the In vitro Maturation of Mouse Oocytes after Mechanical Stimulation by Hydrostatic Pressure

Background: Oocyte maturation is usually defined as the period of progression from the first to the second meiotic arrest and involves coordinated nuclear and cytoplasmic modification. Ca2+ signals are known as important regulators of oocyte maturation. Extracellular Ca2+ is important for first polar body formation, normal meiosis kinetics, and for preserving the chromosome and spindle configur...

Force-Induced Unfolding of Fibronectin in the Extracellular Matrix of Living Cells

Whether mechanically unfolded fibronectin (Fn) is present within native extracellular matrix fibrils is controversial. Fn extensibility under the influence of cell traction forces has been proposed to originate either from the force-induced lengthening of an initially compact, folded quaternary structure as is found in solution (quaternary structure model, where the dimeric arms of Fn cross eac...
